Thymic changes as a contributing factor in the increased susceptibility of old Albino Oxford rats to EAE development

AB  - The study was aimed to examine putative contribution of thymic involution to ageing-associated increase in susceptibility of Albino Oxford (AO) rats to the development of clinical EAE, and vice versa influence of the disease on the progression of thymic involution. To this end we examined (i) the parameters of thymocyte negative selection efficacy, the thymic generation of CD4+CD25+Foxp3+ T regulatory cells (Tregs) and thymic capacity to instruct/predetermine IL-17-producing T-cell differentiation, and thymopietic efficacy-associated accumulation of “inflammescent” cytotoxic CD28- T cells in the periphery, and (ii) the key underlying mechanisms in young and old non-immunised AO rats and their counterparts immunised for EAE (on the 16th day post-immunisation when the disease in old rats reached the plateau) using flow cytometry analysis and/or RT-qPCR. It was found that thymic involution impairs: (i) the efficacy of negative selection (by affecting thymocyte expression of CD90, negative regulator of selection threshold and the expression of thymic stromal cell integrity factors) and (ii) Treg generation (by diminishing expression of cytokines supporting their differentiation/maturation). Additionally, the results suggest that thymic involution facilitates CD8+ T-cell differentiation into IL-17-producing cells (previously linked to the development of clinical EAE in old AO rats). Furthermore, they confirmed that ageing-related decrease in thymic T-cell output (as indicated by diminished frequency of recent thymic emigrants in peripheral blood) resulted in the accumulation of CD28- T cells in peripheral blood and, upon immunisation, in the target organ. On the other hand, the development of EAE (most likely by increasing circulatory levels of proinflammatory cytokines) contributed to the decline in thymic output of T cells, including Tregs, and thereby to the progression/maintenance of clinical EAE. Thus, in AO rats thymic involution via multi-layered mechanisms may favour the development of clinically manifested autoimmunity, which, in turn, precipitates the thymus atrophy.

Adrenoceptors as potential target for add-on immunomodulatory therapy in multiple sclerosis

AB  - This review summarizes recent findings related to the role of the sympathetic nervous system (SNS) in pathogenesis of multiple sclerosis (MS) and its commonly used experimental model – experimental autoimmune encephalomyelitis (EAE). They indicate that noradrenaline, the key end-point mediator of the SNS, acting through β-adrenoceptor, has a contributory role in the early stages of MS/EAE development. This stage is characterized by the SNS hyperactivity (increased release of noradrenaline) reflecting the net effect of different factors, such as the disease-associated inflammation, stress, vitamin D hypovitaminosis, Epstein-Barr virus infection and dysbiosis. Thus, the administration of propranolol, a non-selective β-adrenoceptor blocker, readily crossing the blood-brain barrier, to experimental rats before the autoimmune challenge and in the early (preclinical/prodromal) phase of the disease mitigates EAE severity. This phenomenon has been ascribed to the alleviation of neuroinflammation (due to attenuation of primarily microglial activation/proinflammatory functions) and the diminution of the magnitude of the primary CD4+ T-cell autoimmune response (the effect associated with impaired autoantigen uptake by antigen presenting cells and their migration into draining lymph nodes). The former is partly related to breaking of the catecholamine-dependent self-amplifying microglial feed-forward loop and the positive feedback loop between microglia and the SNS, leading to down-regulation of the SNS hyperactivity and its enhancing influence on microglial activation/proinflammatory functions and the magnitude of autoimmune response. The effects of propranolol are shown to be more prominent in male EAE animals, the phenomenon important as males (like men) are likely to develop clinically more severe disease. Thus, these findings could serve as a firm scientific background for formulation of a new sex-specific immune-intervention strategy for the early phases of MS (characterized by the SNS hyperactivity) exploiting anti-(neuro)inflammatory and immunomodulatory properties of propranolol and other relatively cheap and safe adrenergic drugs with similar therapeutic profile.

Epidemiological Predictors of Positive SARS-CoV-2 Polymerase Chain Reaction Test in Three Cohorts: Hospitalized Patients, Healthcare Workers, and Military Population, Serbia, 2020

AB  - Background: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and its resulting coronavirus disease 2019 (COVID-19) has caused a fast-moving pandemic. Diagnostic testing, aimed to identify patients infected with SARS-CoV-2, plays a key role in controlling the COVID-19 pandemic in different populations. (2) Methods: This retrospective cohort study aimed to investigate predictors associated with positive polymerase chain reaction (PCR) SARS-CoV-2 test results in hospitalized patients, healthcare workers (HCWs), and military personnel (MP) during 2020, before the widespread availability of COVID-19 vaccines. Persons with a positive test result were compared with persons with a negative test result in three cohorts during the study period. (3) Results: A total of 6912 respondents were tested, and 1334 (19.3%) of them had positive PCR SARS-CoV-2 test results. Contact with a known COVID-19 case within 14 days (p < 0.001; OR: 1.48; 95% CI: 1.25–1.76), fever (p < 0.001; OR: 3.66; 95% CI: 3.04–4.41), cough (p < 0.001; OR: 1.91; 95% CI: 1.59–2.30), headache (p = 0.028; OR: 1.24; 95% CI: 1.02–1.50), and myalgia/arthralgia (p < 0.001; OR: 1.99; 95% CI: 1.65–2.42) were independently associated with positive PCR SARS-CoV-2 test results in the cohort of MP. Furthermore, fever (p < 0.001; OR: 2.75; 95% CI: 1.83–4.13), cough (p < 0.001; OR: 2.04; 95% CI: 1.32–3.13), headache (p = 0.008; OR: 1.76; 95% CI: 1.15–2.68), and myalgia/arthralgia (p = 0.039; OR: 1.58; 95% CI: 1.02–2.45) were independently associated with positive PCR SARS-CoV-2 test results in the cohort of HCWs. Moreover, independent predictors of positive PCR SARS-CoV-2 test results in hospitalized patients were contact with a known COVID-19 case within 14 days (p < 0.001; OR: 2.56; 95% CI: 1.71–3.83), fever (p < 0.001; OR: 1.89; 95% CI: 1.38–2.59), pneumonia (p = 0.041; OR: 1.45; 95% CI: 1.01–2.09), and neurological diseases (p = 0.009; OR: 0.375; 95% CI: 0.18–0.78). (4) Conclusions: According to data gathered from cohorts of hospitalized patients, HCWs, and MP, before the widespread availability of COVID-19 vaccines in Serbia, we can conclude that predictors of positive PCR SARS-CoV-2 test results in MP and HCWs were similar. Accurate estimates of COVID-19 in different population groups are important for health authorities.

AB  - Diazinon is an organophosphate pesticide (OP) that has significant potential for accidental and intentional poisoning of wildlife, domestic animals and humans. The aim of the study is to investigate the correlation between cholinesterase activity and oxidative stress parameters in liver and diaphragm by continuous monitoring as a function of time during prolonged use of diazinon. Wistar rats were treated orally with diazinon (55 mg/kg/day): 7, 14, 21 and 28 days. At the end of each period, blood, liver and diaphragm were collected to examine cholinesterase activity and enzymatic/non-enzymatic oxidative stress parameters: superoxide dismutase 1 (SOD1), catalase (CAT), thiobarbituric acid substances (TBARS), protein carbonyl groups. In all four time periods, there was a significant change in acetylcholinesterase (AChE) in erythrocytes and butyrylcholinesterase (BuChE) in blood plasma, CAT in liver and diaphragm and SOD1 in diaphragm. Parameters significantly altered during the cholinergic crisis included: cholinesterases and TBARS in liver and diaphragm and partially SOD1 in liver. Protein carbonyl groups in liver and diaphragm were significantly altered outside the cholinergic crisis. In the liver, there was a very strong negative correlation between BuChE and TBARS in all four time periods and BuChE and CAT on day 7. In the diaphragm, a very strong negative correlation was found between AChE and TBARS at days 7 and 14, and a very strong positive correlation between AChE and SOD1 at days 14, 21 and 28. A better understanding of the relationship between cholinergic overstimulation and oxidative stress may help to better assess health status in prolonged OPs intoxication.

AB  - Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease, affecting motor neurons, which is characterized by the progressive loss of motor function. Sufficient evidence has been gathered, either in the form of experimental results from animal disease models or from the assessment of affected human subjects, which implicates metabolic changes in the course of ALS. Hypermetabolism, abnormal glucose tolerance and dyslipidaemia were reported to have high incidence in ALS patients. In fact, according to published studies, BMI is significantly and inversely associated with ALS progression. Here, we discuss basic principles of fatty acid metabolism, composition and the existing knowledge of metabolic and fatty acid composition changes in ALS patients. Literature data suggest that ALS patients have unfavorable changes in the fatty acid profile, especially because of the low level of long-chain polyunsaturated fatty acids. Nutritional interventions and supplementation with various foods or food supplements are currently not showing conclusive results, implying there is more to learn about potential ways to modify this illness.

AB  - The combination of b-oligosaccharides from enzymatically hydrolysed barley b-glucan has attracted interest recently due to its positive effects on human health. This study aimed to assess the impact of the
endo-b-1,3-1,4-glucanase enzyme from Bacillus subtilis 168 on improving the nutritional and bioactive
properties of barley b-glucan. A new procedure for the isolation of b-glucan was developed, at a lower
temperature (45 °C), enabling purity from starch contamination, without affecting the yield (6 g b-glucan
from 100 g of barley flour). The endo-b-1,3-1,4-glucanase is cloned into E. coli pQE_Ek enables the high
production and purification (82% yield, 1.8 mg mL 1 and 440 U mg 1
) of an enzyme identical to the
natural one (25.5 kDa). The enzymatic reaction showed high efficiency of b-glucan degradation by recombinant enzyme, giving a mixture of products (of which 3-O-b-cellobiosyl-D-glucose and 3-O-b-cellotriosylD-glucose are the most abundant), the reduction of viscosity (17%) and increase in antioxidant capacities
by 15.2%, 30.9% and 44.0% assessed by ABTS, DPPH and ORAC, respectively. These results indicate
the possible application of endo-b-1,3-1,4-glucanase enzyme in improving the properties of barley bglucan used as functional foods.

AB  - α-Amylase from the thermophilic bacterial strain Anoxybacillus vranjensis ST4 (AVA) was cloned into the pMALc5HisEk expression vector and successfully expressed and purified from the Escherichia coli ER2523 host strain. AVA belongs to the GH13_5 subfamily of glycoside hydrolases and has 7 conserved sequence regions (CSRs) distributed in three distinct domains (A, B, C). In addition, there is a starch binding domain (SBD) from the CBM20 family of carbohydrate binding modules (CBMs). AVA is a monomer of 66 kDa that achieves maximum activity at 60–80 °C and is active and stable over a wide pH range (4.0–9.0). AVA retained 50 % of its activity after 31 h of incubation at 60 °C and was resistant to a large number of denaturing agents. It hydrolyzed starch granules very efficiently, releasing maltose, maltotriose and maltopentaose as the main products. The hydrolysis rates of raw corn, wheat, horseradish, and potato starch, at a concentration of 10 %, were 87.8, 85.9, 93.0, and 58 %, respectively, at pH 8.5 over a 3 h period. This study showed that the high level of expression as well as the properties of this highly stable and versatile enzyme show all the prerequisites for successful application in industry.

AB  - Predmet istraživanja ove doktorske disertacije je ispitivanje promena u fenotipu i aktivnosti makrofagaperitonealne šupljine Albino Oxford (AO) i Dark Agouti (DA) pacova tokom razvoja inflamatornogodgovora koji je indukovan delovanjem bakterija mikrobiote creva direktno, intraperitonealnom (i.p.)primenom, ili indirektno, tokom razvoja kolitisa, kao i ispitivanje modulacije ovih promena primenomprobiotske bakterije Lactobacillus rhamnosus (L. rhamnosus).Rezultati su pokazali da i.p. primena umrtvljenih E. coli ili Enterococcus spp. dovodi do izraženijeinflamatorne polarizacije makrofaga peritonealne šupljine DA pacova u odnosu na AO soj, dok i.p.primena L. rhamnosus ima veći potencijal da spreči funkcionalne promene do kojih dovodi in vitrostimulacija komensalnim bakterijama u makrofagima pacova AO soja. Tokom razvoja kolitisamakrofagi DA pacova produkuju više IL-6 u odnosu na AO soj, a značajno manja smrtnost AO pacovaje praćena intenzivnijom produkcijom IL-10 u makrofagima u odgovoru na dodatnu stimulacijukomensalnim bakterijama in vitro i višim nivoom antitela klase IgA specifičnih za E. coli u serumu. Ranipostnatalni oralni tretman bakterijom L. rhamnosus u DA pacovima povećava raznovrsnostBifidobacterium vrsta u fecesu, a nakon indukcije kolitisa u adultnom dobu smanjuje stepen ulceracijekolona, povećava zastupljenost rezidentnih makrofaga peritonealne šupljine i suprimira povećanjaprodukcije NO i IL-10 stimulisana komensalnim bakterijama in vitro.Nalazi ukazuju na značaj genetski determinisane predispozicije ka određenom obrascu aktivacijemakrofaga za promene u fenotipu i aktivnosti tokom razvoja inflamatornog odgovora koji je indukovandelovanjem bakterija mikrobiote creva, kao i za modulaciju ovih promena primenom probiotskebakterije L. rhamnosus.
AB  - The subject of this doctoral dissertation is the study of changes in the phenotype and activity ofperitoneal cavity macrophages from Albino Oxford (AO) and Dark Agouti (DA) rats during thedevelopment of inflammatory response induced by intestinal microbiota directly, intraperitoneally (i.p.),or indirectly, through development of colitis, as well as study of the modulation of these changes usingthe probiotic bacterium Lactobacillus rhamnosus (L. rhamnosus).I.p. application of killed E. coli or Enterococcus spp. leads to a more pronounced inflammatorypolarization of the peritoneal cavity macrophages from DA rats compared the AO strain, while i.p.application of L. rhamnosus has a greater potential to prevent functional changes caused by in vitrostimulation with commensal bacteria in AO rat macrophages. During the development of colitis, DArat macrophages produce more IL-6 than the AO strain, and significantly lower AO rat mortality wasaccompanied by more intense IL-10 production in macrophages in response to additional stimulationwith commensal bacteria in vitro and higher levels of IgA-class antibodies specific to E. coli in the serum.Early postnatal oral treatment with L. rhamnosus in DA rats increased the diversity of Bifidobacteriumspecies in feces, and after induction of colitis in adulthood reduced the degree of colonic ulceration,increased the presence of resident peritoneal cavity macrophages and suppressed the increasedproduction of NO and IL-10 stimulated by commensal bacteria .The findings indicate the importance of genetically determined predisposition to a certain pattern ofmacrophage activation for changes in phenotype and activity during the development of inflammatoryresponse induced by intestinal microbiota, as well as for modulation of these changes using probioticbacterium L. rhamnosus.

abstract = "Predmet istraživanja ove doktorske disertacije je ispitivanje promena u fenotipu i aktivnosti makrofagaperitonealne šupljine Albino Oxford (AO) i Dark Agouti (DA) pacova tokom razvoja inflamatornogodgovora koji je indukovan delovanjem bakterija mikrobiote creva direktno, intraperitonealnom (i.p.)primenom, ili indirektno, tokom razvoja kolitisa, kao i ispitivanje modulacije ovih promena primenomprobiotske bakterije Lactobacillus rhamnosus (L. rhamnosus).Rezultati su pokazali da i.p. primena umrtvljenih E. coli ili Enterococcus spp. dovodi do izraženijeinflamatorne polarizacije makrofaga peritonealne šupljine DA pacova u odnosu na AO soj, dok i.p.primena L. rhamnosus ima veći potencijal da spreči funkcionalne promene do kojih dovodi in vitrostimulacija komensalnim bakterijama u makrofagima pacova AO soja. Tokom razvoja kolitisamakrofagi DA pacova produkuju više IL-6 u odnosu na AO soj, a značajno manja smrtnost AO pacovaje praćena intenzivnijom produkcijom IL-10 u makrofagima u odgovoru na dodatnu stimulacijukomensalnim bakterijama in vitro i višim nivoom antitela klase IgA specifičnih za E. coli u serumu. Ranipostnatalni oralni tretman bakterijom L. rhamnosus u DA pacovima povećava raznovrsnostBifidobacterium vrsta u fecesu, a nakon indukcije kolitisa u adultnom dobu smanjuje stepen ulceracijekolona, povećava zastupljenost rezidentnih makrofaga peritonealne šupljine i suprimira povećanjaprodukcije NO i IL-10 stimulisana komensalnim bakterijama in vitro.Nalazi ukazuju na značaj genetski determinisane predispozicije ka određenom obrascu aktivacijemakrofaga za promene u fenotipu i aktivnosti tokom razvoja inflamatornog odgovora koji je indukovandelovanjem bakterija mikrobiote creva, kao i za modulaciju ovih promena primenom probiotskebakterije L. rhamnosus., The subject of this doctoral dissertation is the study of changes in the phenotype and activity ofperitoneal cavity macrophages from Albino Oxford (AO) and Dark Agouti (DA) rats during thedevelopment of inflammatory response induced by intestinal microbiota directly, intraperitoneally (i.p.),or indirectly, through development of colitis, as well as study of the modulation of these changes usingthe probiotic bacterium Lactobacillus rhamnosus (L. rhamnosus).I.p. application of killed E. coli or Enterococcus spp. leads to a more pronounced inflammatorypolarization of the peritoneal cavity macrophages from DA rats compared the AO strain, while i.p.application of L. rhamnosus has a greater potential to prevent functional changes caused by in vitrostimulation with commensal bacteria in AO rat macrophages. During the development of colitis, DArat macrophages produce more IL-6 than the AO strain, and significantly lower AO rat mortality wasaccompanied by more intense IL-10 production in macrophages in response to additional stimulationwith commensal bacteria in vitro and higher levels of IgA-class antibodies specific to E. coli in the serum.Early postnatal oral treatment with L. rhamnosus in DA rats increased the diversity of Bifidobacteriumspecies in feces, and after induction of colitis in adulthood reduced the degree of colonic ulceration,increased the presence of resident peritoneal cavity macrophages and suppressed the increasedproduction of NO and IL-10 stimulated by commensal bacteria .The findings indicate the importance of genetically determined predisposition to a certain pattern ofmacrophage activation for changes in phenotype and activity during the development of inflammatoryresponse induced by intestinal microbiota, as well as for modulation of these changes using probioticbacterium L. rhamnosus.",

AB  - Our previous studies showed more severe experimental autoimmune encephalomyelitis (EAE) in male compared with female adult rats, and moderating effect of propranolol-induced β-adrenoceptor blockade on EAE in females, the effect associated with transcriptional stimulation of Nrf2/HO-1 axis in spinal cord microglia. This study examined putative sexual dimor- phism in propranolol action on EAE severity. Propranolol treatment beginning from the onset of clinical EAE mitigated EAE severity in rats of both sexes, but to a greater extent in males exhibiting higher noradrenaline levels and myeloid cell β 2 -adrenoceptor expression in spinal cord. This correlated with more prominent stimulatory effects of propranolol not only on CX3CL1/CX3CR1/Nrf2/HO-1 cascade, but also on Stat3/Socs3 signaling axis in spinal cord microglia/myeloid cells (mirrored in the decreased Stat3 and the increased Socs3 expression) from male rats compared with their female counterparts. Propranolol diminished the frequency of activated cells among microglia, increased their phagocyting/endocyting capacity, and shifted cytokine secretory profile of microglia/blood-borne myeloid cells towards an anti-inflammatory/neuroprotective phenotype. Additionally, it downregulated the expression of chemokines (CCL2, CCL19/21) driving T-cell/monocyte traf- ficking into spinal cord. Consequently, in propranolol-treated rats fewer activated CD4+ T cells and IL-17+ T cells, including CD4+IL17+ cells coexpressing IFN-γ/GM-CSF, were recovered from spinal cord of propranolol-treated rats compared with sex-matched saline-injected controls. All the effects of propranolol were more prominent in males. The study as a whole disclosed that sexual dimorphism in multiple molecular mechanisms implicated in EAE development may be responsible for greater severity of EAE in male rats and sexually dimorphic action of substances affecting them.

AB  - S. pneumoniae is an important human pathogen which has a polysaccharide capsule with virulent properties. This work aims to estimate the titres of S. pneumoniae specific IgG and IgA isotypes, with respect to age and sex. An in-house whole bacterial cell ELISA was used for the determination of relative levels and endpoint titres of IgG subclasses and IgA1 subclass specific for S. pneumoniae serogroup 1, and to quantify specific IgG1 and IgG2 levels. Significantly lower anti-pneumococcus IgG1 titres were found in older individuals, which was more pronounced in men. Lower IgG2 titres were detected in men over 50 years of age, in comparison to women under 50 years of age. The levels of IgG3 and IgG4 did not differ between different sex and age groups. Lower IgA1 levels were detected in male individuals in both age groups in comparison to females under 50 years of age. The levels of IgG1 showed a moderate correlation with IgG4 in younger individuals of both sexes (r = 0.61 in men and 0.63 in women) which was not noted in the older age group. We highlight the deficiency in humoral immunity in older people, especially male and suggest immunization of this population with pneumococcal vaccines.

AB  - The surface of microorganisms is covered with carbohydrates, which makes them unique, self-sustaining glycan probes. Lectins are able to bind to these probes, and this interaction can be exploited for selecting microorganisms or novel lectins. To examine lectin-microorganism interactions, we have previously developed an enzyme-linked lectin sorbent assay (ELLSA) with whole bacterial cells. To further test the validity of this methodology, here we compare it with flow cytometry. For this purpose, we used biotinylated recombinantly produced lectin from Musa acuminata (BanLec), this lectin’s recombinantly produced chimera with green fluorescent protein (BanLec-eGFP) and a lectin from Ricinus communis (RCA120), both biotinylated and FITC labeled. Parallel testing showed equivalent results for the two methods, in terms of the presence or absence of binding, with signal intensity yielding high Pearson correlation coefficient of 0.8 for BanLec and 0.95 for RCA120. The ELLSA method demonstrated multiple advantages, such as reliability and convenience for high-throughput analysis; it also required less lectin and yielded more consistent results. As such, ELLSA proved to be a useful tool for profiling microbial glycan structures or testing novel lectins.

AB  - Aims Ageing affects N-methyl-D-aspartate receptors (NMDARs), their expression and function in neuronal and non-neuronal
cells. Contribution of NMDARs to pathogenesis of experimental autoimmune encephalomyelitis (EAE) has been investigated
but further study is still needed. The aim of this study was to determine whether ageing affects the role of NMDARs in EAE.
Methods Memantine, a non-competitive NMDAR antagonist which limits pathological activity of NMDARs while sparing
normal synaptic activity, was administered orally from day 7 after immunization to 3- and 24-month-old female Dark Agouti
rats. The animals were sacrificed at the peak of the disease. Spinal cord mononuclear cells were analyzed by flow cytometry.
Brain tissue was collected for biochemical analysis of redox status and RT-qPCR. Results Semiprophylactic administration of
memantine ameliorated clinical disease course, with greater effect in aged rats. Memantine reduced the number, frequency,
and reactivation of CD4+ T lymphocytes and increased the relative percentage of CX3CR1-expressing microglia in spinal
cord, but to a greater extent in aged rats. Additionally, analysis of brain redox status parameters showed that memantine was
more effective in reducing superoxide anion radical, malondialdehyde and advanced oxidation protein products in aged rats
than in young ones. In accordance with previous findings, NMDAR inhibition by memantine decreased NADPH oxidase and
IL-1β expression and increased the nuclear factor erythroid 2-related factor 2 and heme oxygenase-1 expression, to a greater
extent in aged rats. Conclusions The involvement of NMDARs in the pathogenesis of EAE was age-dependent, being more
pronounced in aged than in young rats.

AB  - BACKGROUND A variety of commensal bacterial taxa elicit serum IgA responses resulting in protection against polymicrobial sepsis, whereas anti-commensal IgG may have deleterious role in gastrointestinal and systemic inflammation. OBJECTIVES The aim was to determine the systemic levels of specific antibodies directed to autologous E.coli in rats of Albino Oxford (AO) and Dark Agouti (DA) rat strains during colitis. METHODS Rats were intrarectally injected with ethanol or trinitrobenzenesulfonic acid (TNBS, 10 or 40mg/kg) whereas controls received saline in the same manner. Sera were tested for the level of anti-E.coli antibodies of IgG1, IgG2a, IgG2b and IgA classes by ELISA. RESULTS Both rat strains developed colitis, but the degree of colon necrosis and hyperemia were slightly greater in DA than in AO rats and the survival rate was significantly lower in DA relative to AO rats. Among saline-treated controls, the levels of IgG1, IgG2a, and IgG2b anti-E.coli antibodies were comparable between rat strains, whereas the amounts of IgA were significantly higher in DA compared to AO rats. Development of colitis significantly increased the amount of anti-E.coli antibodies of IgA and IgG2a classes in sera of AO rats, and those of IgG2a and IgG2b classes in sera of DA rats. Hence, mostly beneficial role of IgA and probably deleterious role of IgG2b antibodies directed to commensal E.coli during colitis may be suggested, whereas the role of anti-E.coli antibodies of IgG2a classes remains intriguing.

AB  - BACKGROUND Deregulation of the immune response to microbiota or pathogens, and increased intestinal permeability have been proposed as disease-driving mechanisms in colitis. Since peritoneal macrophages guard the sterility of peritoneal cavity from bacterial leakage from the gut, it is plausible to assume that peritoneal macrophages are involved in colitis development. OBJECTIVES The objective was to investigate changes in the composition of peritoneal cells and their response to stimulation by selected gut microbiota during colitis. METHODS Seven days following induction of colitis with intrarectal instillation of ethanol or trinitrobenzenesulfonic acid (TNBS, 10mg/kg or 40mg/kg), peritoneal cells of Dark Agouti (DA) rats were isolated and subjected to flow cytometry. The amount of IL-6 and TNF-α produced by adherent cells was determined by ELISA following in vitro stimulation with LPS and commensal E.coli and Enteroccocus spp.
RESULTS
Instillation of ethanol or TNBS (10 and 40 mg/kg) increased the proportion of CD11bintCD4low monocytes
and decreased the proportion of resident CD163+MHCIIIo macrophages and CD163-MHCIIhi macrophage/dendritic cells.
In vitro treatment with Enterococcus spp. was superior over LPS and E.coli in increasing macrophage TNF-α release in all
but saline-injected control rats. In vitro treatment with E.coli exceeded the level of LPS stimulation in
inducing macrophage IL-6 release in saline- and ethanol-injected rats. It may be concluded that changes in the
composition of peritoneal cells during colitis and, subsequently, their selectively altered response to gut commensals
may perpetuate or modulate inflammation during disease development

AB  - Increased interest in microbiota calls for the thorough analysis of antibody reactivity to different microorganisms. As salivary IgA represents the first line of defence against microorganisms contacting mucosal surfaces, we explored the binding and specificity of salivary IgA by testing the binding of purified, FITC-labelled salivary IgA to different microorganisms in flow cytometry and conclude that this kind of analysis enables the differentiation of species/strains with high IgA binding capacity, which should be corroborated on a larger sample size. Further we compare, with in-house ELISA, the binding of polyclonal salivary IgA with the binding of polyclonal serum IgA from the same individuals to whole microbial cells and to purified microbial components. High correlations were obtained in total salivary IgA binding to Lactobacillus rhamnosus and Escherichia coli, very distant bacterial species, as well as to isolated bacterial components (r = .70–.97). The binding of total salivary IgA resembled the binding of both salivary IgA1 and IgA2, with IgA2 predominating. For serum polyclonal IgA repertoire, substantially higher specificity was obtained. Serum IgA binding to E. coli correlated best with serum IgA binding to lipopolysaccharide (r = .86), and serum IgA against L. rhamnosus correlated best with the anti-peptidoglycan IgA levels (r = .88). We have also detected that total serum IgA response is governed by either IgA1 or IgA2 response, depending on the nature of the antigen/s. We conclude that steady state salivary IgA repertoire, unlike serum IgA repertoire, consists of polyreactive antibodies with innate specificity, questioning its capacity to select resident microbiota.

AB  - Background: Large-scale epidemics of haemorrhagic fever with renal syndrome (HFRS) have been reported mostly in Asia and Europe, with around 100,000 people affected each year. In the Southeast Europe, Balkan region, HFRS is endemic disease with approximately 100 cases per year. Our aim was to describe epidemiological characteristics of HFRS in five Western Balkan (WB) countries and to describe correlation between HFRS incidence and major meteorological event that hit the area in May 2014. Methods: National surveillance data of HFRS from Bosnia and Herzegovina, Croatia, Montenegro, North Macedonia and Serbia obtained from 1 January 2006 to 31 December 2015 were collected and analysed. Results: In a 10-year period, a total of 1,065 HFRS patients were reported in five WB countries. Cumulative incidence rate ranged from 0.05 to 15.80 per 100.000 inhabitants (in North Macedonia and Montenegro respectively). Increasing number of HFRS cases was reported with a peak incidence in three specific years (2008, 2012, and 2014). Average incidence for the entire area was higher in males than females (5.63 and 1.90 per 100.000 inhabitants respectively). Summer was the season with the highest number of cases and an average incidence rate of 1.74/100.000 inhabitants across 10-year period. Haemorrhagic fever with renal syndrome incidence was significantly increased (7.91/100.000 inhabitants) in 2014, when a few months earlier, severe floods affected several WB countries. A strong significant negative correlation (r = −.84, p < .01) between the monthly incidence of HFRS and the number of months after May's floods was demonstrated for the total area of WB. Conclusion: Our findings demonstrate that the HFRS incidence had similar distribution (general, age, sex and seasonality) across majority of the included countries. Summer was the season with the highest recorded incidence. Common epidemic years were detected in all observed countries as well as a negative correlation between the monthly incidence of HFRS and the number of months after May's cyclone.

AB  - Since 2017, the incidence rate of rubella in the Russian Federation has been below 1 case per million total population. In addition, no circulation of endemic strains of the rubella virus is recorded evidencing about achieving infection elimination phase. In modern conditions, it is important to constantly monitor the level of herd immunity to the rubella virus to identify epidemically significant population groups, especially in countries lacking rubella vaccination or featured with insufficient disease control. Purpose: to study herd immunity to the rubella virus in selected countries in Eurasia and Africa. Materials and methods. Between 2018 and 2021, 15,594 samples of blood sera were tested for IgG and IgM antibodies to the rubella virus from subjects of different ages obtained from regional measles and rubella surveillance centers in the Northwestern Federal District (NWFD) of the Russian Federation, the Republic of Serbia, South Vietnam, and the Republic of Guinea. The “Anti-Rubella Virus ELISA (IgМ)” and “Anti-Rubella Virus ELISA (IgG)” (Euroimmun, Germany) test kits were used. Statistical data processing was carried out using the MS Excel, Prizm 5.0 (GraphPad Software Inc.), and Statistica 8.0 (StatSoft Inc.) software package. Results. During the observation period (2018–2020) the population seroprevalence of the to the rubella virus in the NWFD of the Russian Federation was 96.6–97.7% and fluctuated slightly both in separate years and among individual age groups evidencing about high coverage of rubella vaccination. In the Republic of Serbia conducting two-fold immunization against rubella the overall seroprevalence rate was lower than in the Russian Federation and comprising 86.8%. The minimum number of IgGpositive sera was recorded in the 2–4-year-old age group pointing to the shortcomings of routine vaccination. In South Vietnam, children aged 1–3 years (41.9%) predominated among those recovering from rubella, i.e. the age cohort that should be protected by vaccination at the age of 18 months. No rubella vaccination is carried out in Guinea. The total proportion of seropositive individuals was 75%; herd immunity to the rubella virus was established mainly among children and adolescents, reaching 90% only in the older age group. 30% of unprotected subjects of the most active reproductive age were identified among the females surveyed in Guinea. Conclusion. Insufficient herd immunity to the rubella virus, identified in a number of countries, may contribute to the maintenance of the infectious process and the spread of infection. Globalization contributes to the virus importation into regions being at the stage of measles and rubella elimination. The results obtained suggest about a need to continue efforts aimed at maintaining epidemiological wellbeing regarding rubella in diverse countries of the world.
AB  - Начиная с 2017 г. в Российской Федерации показатель заболеваемости краснухой находится на уровне ниже 1 случая на 1 млн населения. Также отсутствует циркуляция эндемичных штаммов вируса краснухи. Это свидетельствует о достижении фазы элиминации инфекции. В современных условиях важным является постоянный мониторинг уровня коллективного иммунитета к вирусу краснухи для выявления эпидемически значимых групп населения, особенно в странах, где вакцинация против краснухи не проводится или контроль недостаточен. Цель исследования: изучение коллективного иммунитета к вирусу краснухи в ряде стран Евразии и Африки. Материалы и методы. В период с 2017 по 2021 г. на IgG- и IgM-антитела к вирусу краснухи исследовано 15 594 образца сывороток крови лиц разного возраста, полученные из региональных центров по надзору за корью и краснухой в СЗФО РФ, Республике Сербия, в Южном Вьетнаме, в Гвинейской Республике. Использовали ИФА тест-наборы «Anti-Rubella Virus ELISA IgМ» и «Anti-Rubella Virus ELISA (IgG)» (Euroimmun, Германия). Статистическая обработка результатов проводилась с помощью пакета программ MS Excel, Prizm 5.0 (GraphPadSoftware Inc.), Statistica 8.0 (StatSoft Inc.). Результаты. В СЗФО РФ за период наблюдения серопревалентность населения к вирусу краснухи составляла 96,6–97,7% и колебалась незначительно как по отельным годам, так и среди отдельных возрастных групп, что свидетельствуют о высоком охвате вакцинацией против краснухи. В Республике Сербия общий показатель серопревалентности оказался ниже, чем в РФ, и составил 86,8%. Наименьшее количество IgG-положительных сывороток регистрировали в возрастной группе 2–4 года, что говорит о недостатках плановой вакцинации. В Южном Вьетнаме среди переболевших краснухой преобладали дети в возрасте 1–3 года (41,9%), то есть та группа, которая должна быть максимально защищена плановой прививкой против краснухи в 18 месяцев. В Гвинее специфическая профилактика краснухи не проводится. Общая доля серопозитивных лиц составила 75%, коллективный иммунитет к вирусу краснухи формировался, в основном, среди детей и подростков, достигая 90% лишь в старшей возрастной группе. Среди обследованных женщин Гвинеи выявлено 30% незащищенных лиц наиболее активного репродуктивного возраста. Заключение. Недостаточный уровень коллективного иммунитета к вирусу краснухи, выявленный в ряде стран, может способствовать распространению инфекции, а условия глобализации — импортированию вируса в регионы, находящиеся на этапе элиминации кори и краснухи. Полученные результаты свидетельствуют о необходимости продолжения усилий, направленных на поддержание эпидемиологического благополучия в отношении краснухи в разных странах мира.

Purpose: to study herd immunity to the rubella virus in selected countries in Eurasia and Africa. Materials and methods. Between 2018 and 2021, 15,594 samples of blood sera were tested for IgG and IgM antibodies to the rubella virus from subjects of different ages obtained from regional measles and rubella surveillance centers in the Northwestern Federal District (NWFD) of the Russian Federation, the Republic of Serbia, South Vietnam, and the Republic of Guinea. The “Anti-Rubella Virus ELISA (IgМ)” and “Anti-Rubella Virus ELISA (IgG)” (Euroimmun, Germany) test kits were used. Statistical data processing was carried out using the MS Excel, Prizm 5.0 (GraphPad Software Inc.), and Statistica 8.0 (StatSoft Inc.) software package. Results. During the observation period (2018–2020) the population seroprevalence of the to the rubella virus in the NWFD of the Russian Federation was 96.6–97.7% and fluctuated slightly both in separate years and among individual age groups evidencing about high coverage of rubella vaccination. In the Republic of Serbia conducting two-fold immunization against rubella the overall seroprevalence rate was lower than in the Russian Federation and comprising 86.8%. The minimum number of IgGpositive sera was recorded in the 2–4-year-old age group pointing to the shortcomings of routine vaccination. In South Vietnam, children aged 1–3 years (41.9%) predominated among those recovering from rubella, i.e. the age cohort that should be protected by vaccination at the age of 18 months. No rubella vaccination is carried out in Guinea. The total proportion of seropositive individuals was 75%; herd immunity to the rubella virus was established mainly among children and adolescents, reaching 90% only in the older age group. 30% of unprotected subjects of the most active reproductive age were identified among the females surveyed in Guinea. Conclusion. Insufficient herd immunity to the rubella virus, identified in a number of countries, may contribute to the maintenance of the infectious process and the spread of infection. Globalization contributes to the virus importation into regions being at the stage of measles and rubella elimination. The results obtained suggest about a need to continue efforts aimed at maintaining epidemiological wellbeing regarding rubella in diverse countries of the world., Начиная с 2017 г. в Российской Федерации показатель заболеваемости краснухой находится на уровне ниже 1 случая на 1 млн населения. Также отсутствует циркуляция эндемичных штаммов вируса краснухи. Это свидетельствует о достижении фазы элиминации инфекции. В современных условиях важным является постоянный мониторинг уровня коллективного иммунитета к вирусу краснухи для выявления эпидемически значимых групп населения, особенно в странах, где вакцинация против краснухи не проводится или контроль недостаточен. Цель исследования: изучение коллективного иммунитета к вирусу краснухи в ряде стран Евразии и Африки. Материалы и методы. В период с 2017 по 2021 г. на IgG- и IgM-антитела к вирусу краснухи исследовано 15 594 образца сывороток крови лиц разного возраста, полученные из региональных центров по надзору за корью и краснухой в СЗФО РФ, Республике Сербия, в Южном Вьетнаме, в Гвинейской Республике. Использовали ИФА тест-наборы «Anti-Rubella Virus ELISA IgМ» и «Anti-Rubella Virus ELISA (IgG)» (Euroimmun, Германия). Статистическая обработка результатов проводилась с помощью пакета программ MS Excel, Prizm 5.0 (GraphPadSoftware Inc.), Statistica 8.0 (StatSoft Inc.). Результаты. В СЗФО РФ за период наблюдения серопревалентность населения к вирусу краснухи составляла 96,6–97,7% и колебалась незначительно как по отельным годам, так и среди отдельных возрастных групп, что свидетельствуют о высоком охвате вакцинацией против краснухи. В Республике Сербия общий показатель серопревалентности оказался ниже, чем в РФ, и составил 86,8%. Наименьшее количество IgG-положительных сывороток регистрировали в возрастной группе 2–4 года, что говорит о недостатках плановой вакцинации. В Южном Вьетнаме среди переболевших краснухой преобладали дети в возрасте 1–3 года (41,9%), то есть та группа, которая должна быть максимально защищена плановой прививкой против краснухи в 18 месяцев. В Гвинее специфическая профилактика краснухи не проводится. Общая доля серопозитивных лиц составила 75%, коллективный иммунитет к вирусу краснухи формировался, в основном, среди детей и подростков, достигая 90% лишь в старшей возрастной группе. Среди обследованных женщин Гвинеи выявлено 30% незащищенных лиц наиболее активного репродуктивного возраста. Заключение. Недостаточный уровень коллективного иммунитета к вирусу краснухи, выявленный в ряде стран, может способствовать распространению инфекции, а условия глобализации — импортированию вируса в регионы, находящиеся на этапе элиминации кори и краснухи. Полученные результаты свидетельствуют о необходимости продолжения усилий, направленных на поддержание эпидемиологического благополучия в отношении краснухи в разных странах мира.",

AB  - OBJECTIVE: The aim of this study was to determine any differences in ox- idative stress and inflammation parameters in COPD patients treated with either N-acetyl cys- teine (NAC) alone or with NAC in combination with propolis (NACP). PATIENTS AND METHODS: Forty COPD pa- tients in the exacerbation phase were enrolled into the study and were treated with either NAC (NAC group; n=20) or NACP (NACP group; n=20) twice daily for one month. Redox status was de - termined by measuring superoxide anion (O 2 .– ), advanced oxidation protein products (AOPP), total oxidative status (TOS), prooxidative-anti- oxidant balance (PAB), malondialdehyde (MDA), ischemia modified albumin (IMA) and sever- al other antioxidant markers: superoxide dis- mutase (SOD), paraoxonase 1 (PON1), total sulf- hydryl groups (SHG) and total antioxidant status (TAS). Interleukins 6, 8 and 17 were measured as markers of inflammatory status. RESULTS: Both groups had similar socio-de - mographic and clinical characteristics. Af- ter treatment significantly higher SHG [0.446 (0.395-0.516) vs. 0.292 (0.270-0.325), p<0.001] and significantly lower TOS – 50.6 [49.7-53.4 vs. 73.2 (50.9-84.6), p<0.05] – and IMA [0.650 (0.629-0.682) vs. 0.709 (0.667-0.756), p<0.05] – were found in the NACP group compared to the NAC group. Factorial analysis indicated a larger oxidative stress-inflammatory load in the NAC group after treatment. CONCLUSIONS: From an oxidative stress and inflammatory status perspective, treatment with NACP was more successful than with NAC. The inclusion of propolis into therapy for COPD patients, especially those in the exacerbation phase, could prove beneficial.

AB  - Using the universal primers (UNI16Sfw and UNI16Srev) [1], the representative isolates were identified by 16S rDNA sequencing. Amplification was carried out in a thermal cycler (Applied Biosystems, ThermoFisher Scientific) and DNA fragments were amplified as follows: initial denaturation at 94 °C for 5 min, followed by 30 cycles consisting of denaturation at 94 °C for 1 min, annealing at 55°C for 1 min, and polymerization at 72°C for 1 min, and a final extension at 72°C for 7 min. The expected length was 1549 bp. Aliquots (5 µl) of the amplified products were subjected to electrophoresis in 1% agarose gel (ThermoFisher Scientific) in TAE buffer (40 mM Tris acetate, 1 mM EDTA, pH 8.2). Gels were stained with ethidium bromide (500 ng/mL) and visualized under UV light (BioDoc Analyze). All amplicons were eluted and purified using GeneJet PCR Purification Kit (ThermoScientific) by following the manufacturer`s protocol. The PCR products that we obtained were sequenced by the Macrogen Sequencing Service (Macrogen, Amsterdam, The Netherlands) and analyzed by using BLAST algorithm (http://www.ncbi.nlm.nih.gov/index.html). Selected isolates were identified as follows: isolate B2 - Lacotbacillus reuteri, isolate H10 - Lactobacillus murinus, and isolate J7 - Klebsiella oxytoca [2]. The most numerous colonies belong to isolate B2, hence it was chosen for further characterization as a potential source for exopolysaccharide (EPS) production.

AB  - As one of the most promising groups of microbes, lactic acid bacteria (LAB) can synthesize metabolites that can be used in different industries over the world, mainly in the pharmaceutical, food, and dairy industries. In this study, a novel exopolysaccharide was extracted and isolated from the probiotic strain Lactobacillus reuteri B2, and assessed on biological activity and its possible application as a biosorbent for the removal of Ni2+ ions from contaminated water. New exopolysaccharide was characterized using FTIR, SEM, XRD, NMR, MALDI-TOF MS, and TGA/DTG analysis. Biological assays included antioxidative activity, cytotoxic assay, and adhesion assay of L. reuteri B2 to HT29 cells. Our hypothesis was that if this exopolysaccharide is nontoxic, it can be used as a novel biomaterial for the possible application of the removal of Ni2+ ions from contaminated water. The scavenging effect of nontoxic exopolysaccharide was 76% at 2 mg/mL using ABTS assay, in biological assays, while the removal efficiency of nickel from the aqueous solution was 92.96% in biosorption study. According to these results, this exopolysaccharide can be considered a very promising biomaterial for potential application in different industries, from pharmacy to wastewater treatments.
