AB  - S. pneumoniae is an important human pathogen which has a polysaccharide capsule with virulent properties. This work aims to estimate the titres of S. pneumoniae specific IgG and IgA isotypes, with respect to age and sex. An in-house whole bacterial cell ELISA was used for the determination of relative levels and endpoint titres of IgG subclasses and IgA1 subclass specific for S. pneumoniae serogroup 1, and to quantify specific IgG1 and IgG2 levels. Significantly lower anti-pneumococcus IgG1 titres were found in older individuals, which was more pronounced in men. Lower IgG2 titres were detected in men over 50 years of age, in comparison to women under 50 years of age. The levels of IgG3 and IgG4 did not differ between different sex and age groups. Lower IgA1 levels were detected in male individuals in both age groups in comparison to females under 50 years of age. The levels of IgG1 showed a moderate correlation with IgG4 in younger individuals of both sexes (r = 0.61 in men and 0.63 in women) which was not noted in the older age group. We highlight the deficiency in humoral immunity in older people, especially male and suggest immunization of this population with pneumococcal vaccines.

AB  - This study aimed to analyze an impact of Lactobacillus salivarius BGHO1 and Lactobacillus reuteri BGGO6-55 supplemented to Anemia franciscana cultivation medium on biochemical profile of hatched nauplii. Impacts of different BGHO1:BGGO6-55 ratios (75:25, 50:50 and 25:75) and their total concentrations (250 and 500 g/kg of Anemia cysts) at different timings of bacteria application (pre- and post-hatching) were examined. The effects were evaluated by quantification of naupliar peptide, soluble protein, phospho-, neutral and unsaturated lipid content. Uni and multifactorial analysis of variance (ANOVA) were used to estimate the effects of treatments relative to control and to model factor interactions, respectively Statistical analysis indicated that posthatching application of 75:25 strain ratio at high dose was associated with an increase of neutral lipid amount. Hypothetically, graded response of Artemia nauplii to lactobacilli supplementation was induced with different BGHO1 doses. In contrast to lactobacilli supplementation after Anemia hatching, pre-hatching application of lactobacilli was not associated with an increase of Anemia nutritive profile, though again there was a positive association of lactobacilli concentration with nutrient amount. This implicates interference of lactobacilli with the hatching process. Results presented here facilitate the design of future studies aiming to modify Artemia nutritive profile in accordance with nutritional demands of cultivated fish species.

AB  - Weaning to inert diet in intensively reared pike-perch larvae is confronted with significant fish loss, which prevents successful commercialization of pike-perch production. Achievement of satisfactory feed quality and effective assimilation of nutrients by larval fish is the major challenge in larval production process. Aim of this study was to evaluate whether treatment of live and inert feed with lactobacilli could alleviate growth retardation associated with early weaning of pike-perch reared in recirculating system. Weaning started on 18th day post-hatching (DPH) either as sudden weaning (SW) or by co-administration of Anemia for six days (gradual weaning, GW). Prior to administration to fish, Anemia was treated with Lactobacillus salivarius BGHO1/Lb. reuteri BGGO6-55, while inert feed was treated with Lb. paracasei subsp. paracasei BGHN14/Lb. rhamnosus BGT10. Treatment with lactobacilli slightly raised neutral lipid level in Anemia nauplii, but significantly reduced their content in dry feed. Fish were sampled on the 24th DPH. Survival, morphometric indices, skeleton differentiation, digestive enzyme activity and opportunistic pathogenic bacteria level were assessed in whole fish specimens. GW fish were presented with better survival, body growth and phospholipase A2 (PLA2) activity. Alongside, Vibrio spp. growth was suppressed in these fish and skeleton development was improved, according to Alizarin Red staining and ColIA1/Sparc mRNA expression data. Lactobacilli application in GW fish correlated with an increase of survival, condition factor and growth rate, according to trypsin and chymotrypsin activities, indicating better utilization of dietary proteins for muscle building. In SW fish, lactobacilli elevated chymotrypsin activity, PLA2 to lipase activity ratio and improved survival and ossification, as evident from Alizarin Red staining and ColIA1/Sparc mRNA expression. This indicated improved fatty acid absorption and control of metamorphosis process. Furthermore, lactobacilli suppressed Vibrio spp. growth in SW fish. Aside from demonstrating the ability of lactobacilli to aid weaning in pike-perch larvae, this study indicated that different types of food treatment may direct fish growth in a predictable manner, allowing further cost-effective improvements of larval pike-perch rearing in intensive system.

AB  - The aim of this study was to develop novel hydroxyapatite (HAP)-based bioactive bone replacement materials for segmental osteotomy reconstruction. Customized three-dimensional (3D) bone construct was manufactured from nanohydroxyapatite (nHAP) with poly(lactide-co-glycolide) (PLGA) coating using 3D models derived from the computed tomography (CT) scanning of the rabbit's ulna and gradient 3D printing of the bone substitute mimicking the anatomical shape of the natural bone defect. Engineered construct revealed adequate micro-architectural design for successful bone regeneration having a total porosity of 64% and an average pore size of 256 mu m. Radiography and micro-CT analysis depicted new bone apposition through the whole length of the reconstructed ulna with a small area of non-resorbed construct in the central area of defect. Histological analysis revealed new bone formation with both endochondral and endesmal type of ossification. Immunohistochemistry analysis depicted the presence of bone formation indicators - bone morphogenetic protein (BMP), osteocalcin (OCN) and osteopontin (OPN) within newly formed bone. Manufactured personalized construct acts as a "smart" responsive biomaterial capable of modulating the functionality and potential for the personalized bone reconstruction on a clinically relevant length scale.

Characterisation of the interaction of Pseudomonas putida and Pseudomonas tolaasii with Trichoderma aggressivum

AB  - Green mould disease is caused by Trichoderma aggressivum which colonizes mushroom compost and reduces yield. Two Pseudomonas species are associated with mushroom compost: Pseudomonas putida, which stimulates mushroom pinning, and Pseudomonas tolaasii which has a negative effect on crop production. The aim of this work was to characterize T. aggressivum – Pseudomonas interactions as these may be important factors in the development of green mould disease. P. tolaasii supernatant inhibited growth by 57% but P. putida stimulated growth of T.aggressivum by 44%. Tolaasin production was identified in P. tolaasii cultures with a peak at 96 h. Fluorescent microscopy examination of T. aggressivum hyphae revealed that exposure to P. tolaasii supernatant decreased mycelial formation while increasing the abundance of conidia. Label free proteomic analysis of changes in the abundance of T. aggressivum proteins indicated that exposure to P. tolaasii supernatant lead to an oxidative stress response and catabolic enzyme activation (mitochondrial import inner membrane translocase complex (5.7-fold), oxidoreductase (5.2-fold), glucoamylase (5.1-fold)). Exposure of T. aggressivum to P. putida supernatant lead to an increase in the abundance of proteins associated with growth and development (structural constituents of ribosome (20-fold), H/ACA ribonucleoprotein complex subunit (18-fold), DNA binding and nucleosome assembly protein (5.3-fold), and prefoldin (5-fold)). These results indicate that exposure to P. putida can stimulate the growth of T. aggressivum and this interaction may be an important factor in increasing green mould disease in mushroom crops and so reducing yield.

AB  - Green mould disease of mushroom, Agaricus bisporus,is caused by Trichodermaspecies and can result in substantial crop losses.Label free proteomic analysis of changes in the abundance of A. bisporusproteins following exposure to T. aggressivumsupernatantin vitroindicated increased abundance of proteins associated with an oxidative stress response (zinc ion binding (+6.6 fold); peroxidase activity (5.3-fold); carboxylic ester hydrolase (+2.4 fold); dipeptidase (+3.2 fold); [2Fe-2S] cluster assembly (+3.3 fold)). Proteins that decreased in relative abundance were associated with growth: structural constituent of ribosome, translation (-12 fold), deadenylation-dependent decapping of nuclear-transcribed mRNA (-3.4 fold), and small GTPase mediated signal transduction (-2.6 fold). In vivoanalysis revealed that 10-4 T. aggressivuminoculum decreased the mushroom yield by 29% to 56% and 10-3 T. aggressivuminoculum decreased the mushroom yield by 68% to 100%. Proteins that increased in abundance in A. bisporusin vivofollowing exposure to T. aggressivumindicated an oxidative stress response and included proteins with pyruvate kinase activity (+2.6 fold) and hydrolase activity (+2.1 fold)). The results indicate that exposure of A. bisporusmycelium to T. aggressivum in vitroand in vivoresulted in an oxidative stress response and reduction in growth.

abstract = "Green mould disease of mushroom, Agaricus bisporus,is caused by Trichodermaspecies and can result in substantial crop losses.Label free proteomic analysis of changes in the abundance of A. bisporusproteins following exposure to T. aggressivumsupernatantin vitroindicated increased abundance of proteins associated with an oxidative stress response (zinc ion binding (+6.6 fold); peroxidase activity (5.3-fold); carboxylic ester hydrolase (+2.4 fold); dipeptidase (+3.2 fold); [2Fe-2S] cluster assembly (+3.3 fold)). Proteins that decreased in relative abundance were associated with growth: structural constituent of ribosome, translation (-12 fold), deadenylation-dependent decapping of nuclear-transcribed mRNA (-3.4 fold), and small GTPase mediated signal transduction (-2.6 fold). In vivoanalysis revealed that 10-4 T. aggressivuminoculum decreased the mushroom yield by 29% to 56% and 10-3 T. aggressivuminoculum decreased the mushroom yield by 68% to 100%. Proteins that increased in abundance in A. bisporusin vivofollowing exposure to T. aggressivumindicated an oxidative stress response and included proteins with pyruvate kinase activity (+2.6 fold) and hydrolase activity (+2.1 fold)). The results indicate that exposure of A. bisporusmycelium to T. aggressivum in vitroand in vivoresulted in an oxidative stress response and reduction in growth.",

AB  - The aim of this research was to improve nutritive value of fishmeal-based feed by lactobacilli in order to achieve satisfactory nutrient availability needed to support fish development. Feed was solid-state treated at a laboratory scale with the combination of Lactobacillus paracaseisubsp. paracasei BGHN14 and Lactobacillus rhamnosus BGT10 in different experimental settings, which included the variation of strain ratio, total lactobacilli concentration, percentage of moisture and duration of incubation. Short peptides, soluble proteins, phospho-, neutral and unsaturated lipids were quantified. Differences among treated and control feeds were evaluated by Student t-test, while Gaussian process regression (GPR) modeling was employed to simulate the incubation process and define the optimal treatment combination in the context of overall feed nutritional profile. Treatment duration was shown to be the critical determinant of final outcome, either as single factor or via interaction with strain ratio. Optimal nutrient balance was achieved with 12 h incubation period, 260% moisture, 75:25 and 50:50 BGHN14:BGT10 ratios and 200 mg of lactobacilli per g of dry feed. This study should serve as the basis for large-scale tests which would simulate on-farm production of both fishmeal-based and unconventional, lower cost aquafeed with added value.

AB  - Ocular chlamydial infections with the ocular serovars A, B, Ba, and C of Chlamydia trachomatis represent the world's leading cause of infectious blindness. Carrageenans are naturally occurring, sulfated polysaccharides generally considered safe for food and topical applications. Carrageenans can inhibit infection caused by a variety of viruses and bacteria. To investigate whether iota-carrageenan (I-C) isolated from the red alga Chondrus crispus could prevent ocular chlamydial infection, we assessed if targeted treatment of the conjunctival mucosa with I-C affects chlamydial attachment, entry, and replication in the host cell. Immortalized human conjunctival epithelial cells were treated with I-C prior to C. trachomatis infection and analyzed by flow cytometry and immunofluorescence microscopy. In vivo effects were evaluated in an ocular guinea pig inclusion conjunctivitis model. Ocular pathology was graded daily, and chlamydial clearance was investigated. Our study showed that I-C reduces the infectivity of C. trachomatis in vitro. In vivo results showed a slight reduced ocular pathology and significantly less shedding of infectious elementary bodies by infected animals. Our results indicate that I-C could be a promising agent to reduce the transmission of ocular chlamydial infection and opens perspectives to develop prophylactic approaches to block C. trachomatis entry into the host cell.

AB  - We demonstrated that a recombinant banana lectin (rBanLec), which structural characteristics and physiological impacts highly resemble those reported for its natural counterparts, binds murine peritoneal macrophages and specifically modulates their functional characteristics. By using rBanLec in concentrations ranging from 1 mu g to 10 mu g to stimulate resident (RMs) and thioglycollate-elicited (TGMs) peritoneal macrophages from BALB/c and C57BL/6 mice, we have shown that effects of rBanLec stimulation depend on its concentration but also on the functional status of macrophages and their genetic background. rBanLec, in a positive dose-dependent manner, promotes the proliferation of TGMs from both BALB/c and C57BL/6 mice, while its mitogenic influence on RMs is significantly lower (BALB/c mice) or not detectable (C57BL/6 mice). In all peritoneal macrophages, irrespective of their type and genetic background, rBanLec, in a positive dose dependent manner, enhances the secretion of IL-10. rBanLec stimulation of RMs from both BALB/c and C57BL/6 resulted in a positive dose-dependent promotion of proinflammatory phenotype (enhancement of NO production and IL-12 and TNF alpha secretion, reduction of arginase activity). Positive dose-dependent skewing toward proinflammatory phenotype was also observed in TGMs from C57BL/6 mice. In all peritoneal macrophages, irrespective of their type and genetic background, rBanLec, in a positive dose dependent manner, enhances the secretion of IL-10. rBanLec stimulation of RMs from both BALB/c and C57BL/6 resulted in a positive dose-dependent promotion of proinflammatory phenotype (enhancement of NO production and IL-12 and TNF alpha secretion, reduction of arginase activity). Positive dose-dependent skewing toward proinflammatory phenotype was also observed in TGMs from C57BL/6 mice. AB  - Following infection, the balance between protective immunity and immunopathology often depends on the initial infectious load. Several studies have investigated the effect of infectious dose; however, the mechanism by which infectious dose affects disease outcomes and the development of a protective immune response is not known. The aim of this study was to investigate how the infectious dose modulates the local and systemic humoral and the cellular immune responses during primary ocular chlamydial infection in the guinea pig animal model. Guinea pigs were infected by ocular instillation of a Chlamydophila caviae-containing eye solution in the conjunctival sac in three different doses: 1x10(2), 1x10(4), and 1x10(6) inclusion forming units (IFUs). Ocular pathology, chlamydial clearance, local and systemic C. caviae-specific humoral and cellular immune responses were assessed. All inocula of C. caviae significantly enhanced the local production of C. caviae-specific IgA in tears, but only guinea pigs infected with the higher doses showed significant changes in C. caviae-specific IgA levels in vaginal washes and serum. On complete resolution of infection, the low dose of C. caviae did not alter the ratio of CD4(+) and CD8(+) cells within guinea pigs' submandibular lymph node (SMLN) lymphocytes while the higher doses increased the percentages of CD4(+) and CD8(+) cells within the SMLN lymphocytes. A significant negative correlation between pathology intensity and the percentage of CD4(+) and CD8(+) cells within SMLN lymphocyte pool at selected time points post-infection was recorded for both 1x10(4), and 1x10(6) IFU infected guinea pigs. The relevance of the observed dose-dependent differences on the immune response should be further investigated in repeated ocular chlamydial infections.

AB  - We examined the accessibility of influenza virus and diphtheria-tetanus toxin (DiTe) antigens to the choroid plexus (CP) within the postvaccination period and the expression of CD11b molecules (by immunohistochemistry). Eighteen Dark Agouti (DA) rats were divided into three groups: (i) animals administered with influenza vaccine (Flu), (ii) animals administered with DiTe vaccine (DiTe), and (iii) nontreated (Contr) animals. The serum antibody titers following influenza and diphtheria-tetanus vaccination were detected by the ELISA test. Immunohistochemical staining revealed a great number of viral antigen-positive and CD11b-positive brain cells in Flu rats compared to a very small number of the respective cells in DiTe animals and no staining in the Contr group. DiTe- and Flu-rats showed significant increases in the serum anti-tetanus toxoid and anti-influenza virus antibody levels compared to those in the Contr group. The results obtained attract attention towards the dynamic role of the CP in the immunosurveillance of the CNS. Based on the viral antigen deposits accumulated in the CP, it has been proposed that the latter can play an active role in modulation of the immune response after influenza vaccine immunization.

AB  - This research aimed to test the effects of lactobacilli, applied to cultured pike-perch, either through hydrolyzed OTOHIME fish diet, or through Artemia nauplii, on fish growth, microbiota balance and skeletal development. On the 12th Day Post Hatching (DPH) fish were divided into following treatment groups: two groups received the combination of OTOHIME and nauplii enriched either with Lactobacillus paracasei BGHN14 + Lactobacillus rhamnosus BGTH or with Lactobacillus reuteri BGGO6-55 + Lactobacillus salivarius BGHO1, and one group received OTOHIME hydrolyzed by BGHN14 + BGT10 and non-enriched nauplii. Control group received non-enriched nauplii and non-hydrolyzed OTOHIME. The treatment lasted 14 days and fish were sacrificed on the 26th DPH for the assessment of digestive enzyme activity and microbiota composition. Individual total lengths and individual body weights were recorded at the end of the treatments, on the 26th DPH, and also on the 45th DPH, in parallel with the evaluation of skeletal deformities and fish survival. Our results indicated positive effect of Anemia enriched with BGGO6-55 + BGHO1 on fish growth, skeletal development and trypsin to chymotrypsin activity ratio (T/C), as an indicator of protein digestibility. Hydrolysis of OTOHIME was also associated with better skeletal development, higher T/C values and lower levels of Aeromonas and Mycobacterium spp., which are important fish pathogens. Though additional testing in larger cohort studies is needed, these observations are promising in terms of usage of probiotics for improved environmentally friendly production of pike-perch in Recirculating Aquaculture System (RAS).

AB  - Recombinant banana lectin isoform (rBanLec) attaches specifically to the mucosal surface, crosses the epithelial barrier and then directly affects the immune response in mouse colon. Structural characteristics, specificity and physiological impacts of rBanLec reported until now highly resemble those of its natural counterpart. Here, we demonstrated that a dose dependent stimulation of the colon with rBanLec skewed the immune response towards Th1/Th17 direction and this effect was counterbalanced by the rise in IL-10 production. Qualitative and quantitative characteristics of the established cytokine network were dependent on the applied rBanLec concentration. In addition, rBanLec enhanced local NO production and myeloperoxidase activity and promoted an increase in local IgA and IgG production. Stimulation with rBanLec can be beneficial in prevention of pathologies raised due to inappropriate cell-mediated immune response as well as in prevention of the pathogen invasion via the colon. AB  - Antibodies capable to neutralize tetanus toxin (TeNT) are key factors in protection against tetanus disease. Although antibody-based therapeutics for treatment of tetanus exist on the market its production is tedious. Hence, the tetanus-specific antibodies preparation that could be easily produced in large scale in vitro would be beneficial. Monoclonal antibodies (MAbs) are considered for a long time as a reagent of choice, but the core drawback is how to select a MAb that would be safe in providing efficacious protection. In this study we have investigated the parameters crucial for a single MAb to be assigned as protective. Eight murine MAbs were characterized in vitro for their reactivity toward TeNT and assessed in vivo for protectiveness against TeNT intoxication. Correlation of in vitro and in vivo data has revealed that in vitro selection of MAb that is protective in vivo could be performed by a combination of two assays: the measurement of MAb affinity toward TeNT taking Ka 1 x 10(8) M-1 as a threshold level, and the evaluation of its capability to prevent TeNT-ganglioside interaction. Single MAb could be taken into consideration as a potential therapeutic only if it has a capacity to completely inhibits TeNT-ganglioside complex formation. (C) 2015 Elsevier Ltd. Trichoderma species causing green mould disease of button mushroom [Agaricus bisporus (Lange) Imbach] and their sensitivity to fungicides and biofungicides

AB  - Trichoderma species, the causal agents of green mould disease, induce great losses in button mushroom (Agaricus bisporus) farms. Twenty Trichoderma isolates were collected on 13 Serbian A. bisporus farms and one in Bosnia and Herzegovina during 2006 – 2010. Twelve isolates were classified into five species by standard mycological studies and ITS1/ITS4 sequence analyses, namely Trichoderma atroviride, Trichoderma koningii, Trichoderma virens, Trichoderma aggressivum f. europaeum and Trichoderma harzianum. Eight isolates were not identified to the species level but were shown to be related to T. harzianum. The isolates of T. harzianum exhibited the highest virulence to the harvested A. bisporus pilei and T. virens and T. aggressivum f. europaeum the lowest. Fungicides are widely used to control mushroom diseases although green mould control is encumbered with difficulties. The aims of this study were therefore to research in vitro toxicity of several commercial fungicides to Trichoderma isolates originating from Serbian and Bosnia-Herzegovina farms, and to evaluate the effects of pH and light on their growth. The majority of isolates demonstrated optimal growth at pH 5.0, and the rest at pH 6.0. A few isolates also grew well at pH 7. The weakest mycelial growth was noted at pH 8.0 – 9.0. Generally, light had an inhibitory effect on the growth of tested isolates. The isolates showed the highest susceptibility to prochloraz-Mn, chlorothalonil and carbendazim (ED50 less than 1 mg/L), and were less sensitive to iprodione (ED50 ranged 0.84 – 6.72 mg/L), weakly resistant to thiophanate-methyl (ED50 = 3.75 – 24.13 mg/L), and resistant to trifloxystrobin (ED50 = 10.25 – 178.23 mg/L). Considering the toxicity of fungicides to A. bisporus, prochloraz-Mn and carbendazim showed the best selective toxicity (0.05 and 0.02 respectively), iprodione and chlorothalonil moderate (0.16), and thiophanate-methyl the lowest (1.24), while trifloxystrobin toxicity to A. bisporus was not tested because of its inefficiency against Trichoderma isolates. The best selective toxicity to both pathogen and the host showed prochloraz-Mn and carbendazim with values less than 0.1. Carbendazim was withdrawn from the market and therefore prochloraz-Mn could be recommended for mushroom cultivation in Serbia. Antifungal activity of two biofungicides based on Bacillus subtilis and tea tree oil were tested in vitro to all Trichoderma isolates. B. subtilis was highly toxic to all tested Trichoderma isolates, their ED50 values were below 1.3 mg/L. Tea tree oil did not exhibit a significant antifungal activity (ED50 = 11.9 – 370.8 mg/L). The efficacy of biofungicides was evaluated against T. harzianum in a mushroom growing room, and they were applied alone or in combination with the fungicide at a respective proportion of 20:80%. Prochloraz-Mn showed higher efficacy than both tested biofungicides or their respective mixtures. The biofungicide based on B. subtilis demonstrated greater efficacy in preventing disease symptoms than tea tree oil. B. subtilis combined with the fungicide revealed less antagonism in efficacy against pathogen than tea tree oil.
AB  - Pojedine vrste roda Trichoderma su uzročnici zelene plesni šampinjona (Agaricus bisporus) i nanose velike materijalne štete u gajilištima. Od 2006. do 2010. godine prikupljeno je 20 izolata roda Trichoderma iz 13 gajilišta u Srbiji i jednog iz Bosne i Hercegovine. Dvanaest izolata je klasifikovano na osnovu standardnih mikoloških metoda i analizom ITS1/ITS4 sekvence u pet vrsta: Trichoderma atroviride, Trichoderma koningii, Trichoderma virens, Trichoderma aggressivum f. europaeum i Trichoderma harzianum. Osam izolata nije identifikovano do nivoa vrste ali je pokazano da su filogenetski bliski sa vrstom T. harzianum. Najvišu virulentnost na ubranom šeširu A. bisporus je pokazala vrsta T. harzianum, a najnižu T. virens i T. aggressivum f. europaeum. Većina izolata je najbolje rasla na podlozi koja je imala pH = 5, potom na pH = 6, par izolata na pH = 7, dok je najslabiji rast zabeležen na pH = 8 – 9. Generalno, svetlost je imala inhibitorni efekat na rast izolata. Fungicidi su u širokoj upotrebi u kontroli bolesti šampinjona, iako je kontrola zelene plesni otežana. Stoga, jedan od ciljeva istraživanja je bio da se ispita in vitro toksičnost nekoliko dostupnih komercijalnih fungicida na prikupljene izolate roda Trichoderma. Testirani izolati su bili najosetljivi na prohloraz-Mn, hlorotalonil i karbendazim (EC50  1 mg/L), osetljivi na iprodion (EC50 = 0,84 – 6,72 mg/L), umereno rezistentni na tiofanat-metil (EC50 = 3,75 – 24,13 mg/L) i rezistentni na trifloksistrobin (EC50 = 10,25 – 178,23 mg/L). Proučavajući toksičnost fungicida na A. bisporus, najbolju selektivnu toksičnost su pokazali prohloraz-Mn (0,05) i karbendazim (0,02), iprodion i hlorotalonil umerenu (0,16), tiofanat-metil najnižu (1,24), dok toksičnost trifloksistrobina za A. bisporus nije testirana zbog nezadovoljavajuće toksičnosti za Trichoderma spp. izolate. Najbolju selektivnu toksičnost za patogenu gljivu i gljivu domaćina su pokazali prohloraz-Mn i karbendazim sa vrednostima nižim od 0,1. Pošto se karbendazim povlači sa tržišta, prohloraz-Mn je fungicid koji se može preporučiti za primenu u gajilištima za suzbijanje zelene plesni. Antifungalna aktivnost dva biofungicida, na bazi Bacillus subtilis i ulja čajnog drveta, je testirana in vitro na sve Trichoderma spp. izolate. B. subtilis je bio visoko toksičan za sve testirane izolate (EC50  lt 1,3 mg/L), dok ulje čajnog drveta nije pokazalo značajnu antifungalnu aktivnost (EC50 = 11,9 – 370,8 mg/L). Efikasnost biofungicida protiv vrste T. harzianum je ocenjena u oglednom gajilištu. Biofungicidi su primenjeni zasebno ili u kombinaciji sa fungicidom, prohloraz- Mn, u proporciji 20:80%. Prohloraz-Mn se pokazao efikasnijim od oba biofungicida kao i od kombinovane primene biofungicida i fungicida. Biofungicid na bazi B. subtilis je bio efikasniji od ulja čajnog drveta u pogledu sprečavanja pojave simptoma bolesti. Takođe, B. subtilis je pokazao manji antagonistički efekat u efikasnosti protiv patogena od ulja čajnog drveta kada se primenjuju u kombinaciji sa prohloraz-Mn fungicidom.

abstract = "Trichoderma species, the causal agents of green mould disease, induce great losses in button mushroom (Agaricus bisporus) farms. Twenty Trichoderma isolates were collected on 13 Serbian A. bisporus farms and one in Bosnia and Herzegovina during 2006 – 2010. Twelve isolates were classified into five species by standard mycological studies and ITS1/ITS4 sequence analyses, namely Trichoderma atroviride, Trichoderma koningii, Trichoderma virens, Trichoderma aggressivum f. europaeum and Trichoderma harzianum. Eight isolates were not identified to the species level but were shown to be related to T. harzianum. The isolates of T. harzianum exhibited the highest virulence to the harvested A. bisporus pilei and T. virens and T. aggressivum f. europaeum the lowest. Fungicides are widely used to control mushroom diseases although green mould control is encumbered with difficulties. The aims of this study were therefore to research in vitro toxicity of several commercial fungicides to Trichoderma isolates originating from Serbian and Bosnia-Herzegovina farms, and to evaluate the effects of pH and light on their growth. The majority of isolates demonstrated optimal growth at pH 5.0, and the rest at pH 6.0. A few isolates also grew well at pH 7. The weakest mycelial growth was noted at pH 8.0 – 9.0. Generally, light had an inhibitory effect on the growth of tested isolates. The isolates showed the highest susceptibility to prochloraz-Mn, chlorothalonil and carbendazim (ED50 less than 1 mg/L), and were less sensitive to iprodione (ED50 ranged 0.84 – 6.72 mg/L), weakly resistant to thiophanate-methyl (ED50 = 3.75 – 24.13 mg/L), and resistant to trifloxystrobin (ED50 = 10.25 – 178.23 mg/L). Considering the toxicity of fungicides to A. bisporus, prochloraz-Mn and carbendazim showed the best selective toxicity (0.05 and 0.02 respectively), iprodione and chlorothalonil moderate (0.16), and thiophanate-methyl the lowest (1.24), while trifloxystrobin toxicity to A. bisporus was not tested because of its inefficiency against Trichoderma isolates. The best selective toxicity to both pathogen and the host showed prochloraz-Mn and carbendazim with values less than 0.1. Carbendazim was withdrawn from the market and therefore prochloraz-Mn could be recommended for mushroom cultivation in Serbia. Antifungal activity of two biofungicides based on Bacillus subtilis and tea tree oil were tested in vitro to all Trichoderma isolates. B. subtilis was highly toxic to all tested Trichoderma isolates, their ED50 values were below 1.3 mg/L. Tea tree oil did not exhibit a significant antifungal activity (ED50 = 11.9 – 370.8 mg/L). The efficacy of biofungicides was evaluated against T. harzianum in a mushroom growing room, and they were applied alone or in combination with the fungicide at a respective proportion of 20:80%. Prochloraz-Mn showed higher efficacy than both tested biofungicides or their respective mixtures. The biofungicide based on B. subtilis demonstrated greater efficacy in preventing disease symptoms than tea tree oil. B. subtilis combined with the fungicide revealed less antagonism in efficacy against pathogen than tea tree oil., Pojedine vrste roda Trichoderma su uzročnici zelene plesni šampinjona (Agaricus bisporus) i nanose velike materijalne štete u gajilištima. Od 2006. do 2010. godine prikupljeno je 20 izolata roda Trichoderma iz 13 gajilišta u Srbiji i jednog iz Bosne i Hercegovine. Dvanaest izolata je klasifikovano na osnovu standardnih mikoloških metoda i analizom ITS1/ITS4 sekvence u pet vrsta: Trichoderma atroviride, Trichoderma koningii, Trichoderma virens, Trichoderma aggressivum f. europaeum i Trichoderma harzianum. Osam izolata nije identifikovano do nivoa vrste ali je pokazano da su filogenetski bliski sa vrstom T. harzianum. Najvišu virulentnost na ubranom šeširu A. bisporus je pokazala vrsta T. harzianum, a najnižu T. virens i T. aggressivum f. europaeum. Većina izolata je najbolje rasla na podlozi koja je imala pH = 5, potom na pH = 6, par izolata na pH = 7, dok je najslabiji rast zabeležen na pH = 8 – 9. Generalno, svetlost je imala inhibitorni efekat na rast izolata. Fungicidi su u širokoj upotrebi u kontroli bolesti šampinjona, iako je kontrola zelene plesni otežana. Stoga, jedan od ciljeva istraživanja je bio da se ispita in vitro toksičnost nekoliko dostupnih komercijalnih fungicida na prikupljene izolate roda Trichoderma. Testirani izolati su bili najosetljivi na prohloraz-Mn, hlorotalonil i karbendazim (EC50  1 mg/L), osetljivi na iprodion (EC50 = 0,84 – 6,72 mg/L), umereno rezistentni na tiofanat-metil (EC50 = 3,75 – 24,13 mg/L) i rezistentni na trifloksistrobin (EC50 = 10,25 – 178,23 mg/L). Proučavajući toksičnost fungicida na A. bisporus, najbolju selektivnu toksičnost su pokazali prohloraz-Mn (0,05) i karbendazim (0,02), iprodion i hlorotalonil umerenu (0,16), tiofanat-metil najnižu (1,24), dok toksičnost trifloksistrobina za A. bisporus nije testirana zbog nezadovoljavajuće toksičnosti za Trichoderma spp. izolate. Najbolju selektivnu toksičnost za patogenu gljivu i gljivu domaćina su pokazali prohloraz-Mn i karbendazim sa vrednostima nižim od 0,1. Pošto se karbendazim povlači sa tržišta, prohloraz-Mn je fungicid koji se može preporučiti za primenu u gajilištima za suzbijanje zelene plesni. Antifungalna aktivnost dva biofungicida, na bazi Bacillus subtilis i ulja čajnog drveta, je testirana in vitro na sve Trichoderma spp. izolate. B. subtilis je bio visoko toksičan za sve testirane izolate (EC50  lt 1,3 mg/L), dok ulje čajnog drveta nije pokazalo značajnu antifungalnu aktivnost (EC50 = 11,9 – 370,8 mg/L). Efikasnost biofungicida protiv vrste T. harzianum je ocenjena u oglednom gajilištu. Biofungicidi su primenjeni zasebno ili u kombinaciji sa fungicidom, prohloraz- Mn, u proporciji 20:80%. Prohloraz-Mn se pokazao efikasnijim od oba biofungicida kao i od kombinovane primene biofungicida i fungicida. Biofungicid na bazi B. subtilis je bio efikasniji od ulja čajnog drveta u pogledu sprečavanja pojave simptoma bolesti. Takođe, B. subtilis je pokazao manji antagonistički efekat u efikasnosti protiv patogena od ulja čajnog drveta kada se primenjuju u kombinaciji sa prohloraz-Mn fungicidom.",
